چکیده

The available evidence indicates that formic acid is the precursor of the molecular hydrogen produced as an end product of fermentations effected by Escherichia coli, Aerobacter aerogenes, and related organisms. Formate is rapidly decomposed by these bacteria with the formation of H2 and CO2 in a reaction catalyzed by the enzyme or enzyme-complex designated as "formic hydrogenlyase". One of the major obstacles in elucidating the mechanism of this decomposition has been the inability to obtain the system in the cell-free state. In the present paper the preparation and several properties of a cell-free "formic hydrogenlyase" from E. coli are described.

The inhibition by nitrate of enzyme formation during growth of Escherichia coli.

During an investigation of the effects of nutrients on formic hydrogenlyase activity in Escherichia coli, it was noted that the harvested cells possessed no measurable formic hydrogenlyase activity when grown in a basal medium composed of inorganic salts and glucose and containing NH4NOs as the sole source of nitrogen. This was in agreement with the data reported in a previous communication (Bi...

Enzymes Released from Escherichia Coli with the Aid of a Servall Cell Fractionator.

The release and stability of the enzymes S-adenosylhomocysteine nucleosidase, lysine decarboxylase, arginine decarboxylase, glutamic decarboxylase, formic hydrogenlyase, formic oxidase, and glucose oxidase from Escherichia coli during disruption of the organisms in a Servall-Ribi refrigerated cell fractionator were examined. With the possible exception of arginine decarboxylase, maximal activit...

A study of the hydrogenlyase reaction with systems derived from normal and anaerogenic coli-aerogenes bacteria.

The mechaDism of this process, i. e., the hydrogenlyase reaction, has been the subject of considerable research and speculation over several decades. One fundamental aspect of the reaction which has received particular attention is the question of whether one or more enzymes are required for the conversion. It seems reasonable that definitive and unambiguous evidence relating to this point will...

Efficient Hydrogen-Dependent Carbon Dioxide Reduction by Escherichia coli

Hydrogen-dependent reduction of carbon dioxide to formic acid offers a promising route to greenhouse gas sequestration, carbon abatement technologies, hydrogen transport and storage, and the sustainable generation of renewable chemical feedstocks [1]. The most common approach to performing direct hydrogenation of CO2 to formate is to use chemical catalysts in homogeneous or heterogeneous reacti...

Enhanced hydrogen production from formic acid by formate hydrogen lyase-overexpressing Escherichia coli strains.

Genetic recombination of Escherichia coli in conjunction with process manipulation was employed to elevate the efficiency of hydrogen production in the resultant strain SR13 2 orders of magnitude above that of conventional methods. The formate hydrogen lyase (FHL)-overexpressing strain SR13 was constructed by combining FHL repressor (hycA) inactivation with FHL activator (fhlA) overexpression. ...
